CHAPTER FOUR PROTEOME COMPARISON OF Pseudomonas aeruginosa PLANKTONIC, SURFACE INFLUENCED PLANKTONIC AND BIOFILM POPULATIONS BASED UPON COMPOSITE TWO-DIMENSIONAL ELECTROPHORESIS GELS

چکیده
It has long been recognised that bacteria can switch from planktonic unicellular organisms to sessile multicellular communities known as biofilms (Costerton et al., 1987; 1995). The transition to surface-attached (biofilm) growth is known to result in diverse changes in gene expression, which causes the attaching cells to become phenotypically and metabolically distinct from their planktonic counterparts (Costerton et al., 1999; Davey and O’Toole, 2000; Stewart and Costerton, 2001). Pseudomonas aeruginosa, a ubiquitous environmental gramnegative bacterium, is one of the most studied biofilm-forming organisms and has emerged as a model organism in the study of surfaceand biofilm-induced gene expression.

The use of glass wool as an attachment surface for studying phenotypic changes in Pseudomonas aeruginosa biofilms by two-dimensional gel electrophoresis.
Two-dimensional polyacrylamide gel electrophoresis was used to demonstrate phenotypic differences between Pseudomonas aeruginosa biofilm cells and the planktonic counterpart cells under defined culture conditions. Glass wool was used as a substratum for cell attachment as it affords a large surface-to-volume ratio (1 g with a mean diameter of 15 microns = 1300 cm2), supports the growth of biofi...

Complementary approaches were employed to characterize transitional episodes in Pseudomonas aeruginosa biofilm development using direct observation and whole-cell protein analysis. Microscopy and in situ reporter gene analysis were used to directly observe changes in biofilm physiology and to act as signposts to standardize protein collection for two-dimensional electrophoretic analysis and pro...

متن کاملUse of newly isolated phages for control of Pseudomonas aeruginosa PAO1 and ATCC 10145 biofilms.
Pseudomonas aeruginosa is a relevant opportunistic pathogen involved in nosocomial infections that frequently shows low antibiotic susceptibility. One of its virulence factors is associated with the ability to adhere to surfaces and form virulent biofilms. This work describes the isolation and characterization of lytic phages capable of infecting antibiotic-resistant P. aeruginosa strains. In a...
متن کاملDo Not Fear Commitment: The Initial Transition to a Surface Lifestyle by Pseudomonads
This chapter aims to describe the early stages of biofilm formation, particularly on abiotic surfaces, by focusing on Pseudomonas aeruginosa. Specifically, we will dissect the early steps in the establishment of a multicellular community: (i) translocation to the surface from a free-swimming planktonic lifestyle, (ii) initial or reversible attachment, and finally (iii) irreversible attachment. ...
